Semen/Sperm Detection Methods
Various methods are available for the detection of seminal fluid. Detection of Acid Phosphatase (AP) is a presumptive rather than a confirmatory test because the AP enzyme may be found in bacteria, some vegetables, animals and other human body fluids.

The microscopic identification of sperm cells is conclusive but they may not be present in vasectomized men or those with a low sperm count.
However, the detection of Prostate Specific Antigen (PSA or p30) is a confirmatory test for the presence of semen even in vasectomized or azoospermic individuals. The test is validated for use in the forensic identification of semen and is commonly used by state police crime laboratories in criminal cases. |

Infidelity Testing by IntelliGenetics Seminal fluid released by the male during sexual contact continues to be expelled for several hours after the sexual encounter. Post-intercourse discharge of seminal fluid from the vaginal tract of the women also occurs for several hours. Therefore, following sexual intercourse, semen stains can be found in undergarments, bed sheets, towels, women’s sanitary napkins and other items and surfaces. These facts are the basis for the Infidelity test offered by IntelliGenetics:

Sample Collection & Shipping F Allow the sample to completely air dry; F Cut the article suspected of having a stain or, if preferred, send the entire article.* F Do not store the sample in a plastic bag or air tight container; F Place samples in a paper bag or envelope for overnight shipment.
* Note: If the article is large (bed sheet, blanket, coat etc.) cut out the area in question. Testing is generally destructive to the article. If the entire article is shipped then mark the area in question by circling it with a marker. Articles will not be returned unless requested. A test that is not destructive to the article is available but is less sensitive.
